Octreotide impurities
Octreotide is a somatostatin analogue. Octreotide acetate is a cyclic peptide compound composed of 8 amino acids and is a somatostatin analogue. Octreotide acetate mainly binds with the somatostatin receptor expressed on the surface of tumor cells or other tissue cell membranes with high affinity and specificity to inhibit pituitary growth hormone and the secretion of polypeptide hormones by the gastrointestinal and pancreatic system. Its acetate is used clinically.
Peptide drugs are composed of different amino acids in a certain order. There are many synthetic steps and the impurities generated are also complex. The main impurities include process impurities such as missing peptides, broken peptides and inserted peptides, degradation impurities such as oxidation-reduction, hydrolysis and disulfide bond mismatch, polymer impurities, optical impurities, etc
Octreotide acetate injection is unstable to high temperature, high humidity and strong light, and unstable under accelerated test. Therefore, the impurities of octreotide acetate mainly come from the production and storage process of the preparation, and the production process of the preparation should be further improved and the storage conditions should be strictly controlled. The study of the impurity mass spectra of octreotide acetate and its preparation should be carried out to analyze the factors affecting the generation of impurities, It can purposefully control some impurities and provide reference for the quality control of peptide drugs.
