General Description
Among many coupling agents, TBTU is the most widely used and cost-effective. Urea ionic peptide condensing agent is a multifunctional reagent for activating carboxylic acid. They can combine amino acids with amines in one step without pre activating acids. Their advantages include convenient and practical reaction process, rapid in situ activation, minimal side reactions, commercial availability, stability, solubility in a wide range of solvents including water and easy removal of coupled by-products. Because of the good practicability of urea ionic peptide condensers, the development of their diversified applications has become a new research hotspot.
1. Using TBTU as coupling agent, acetonitrile as solvent, different sulfonic acids and oximes were synthesized in good yields at 50 ℃. The advantages of this method are that the reaction process is simple, the environment is harmless and the post-treatment is simple.
2. Using TBTU as coupling agent, acetonitrile as solvent, carboxylic acid and heterocyclic secondary amines such as indole and imidazole were synthesized in good yields at general temperature. The advantages of this method are that the by-products have little pollution to the environment, the products are easy to separate and the reaction process is simple.
3. A series of urea and carbamate compounds were synthesized with TBTU as coupling agent, acetonitrile as solvent, carboxylic acid and sodium azide, amines or alcohols. This method uses one pot operation, which is not only convenient but also high yield.
